抗非特异性蛋白吸附的聚乙二醇接枝聚乳酸对成骨细胞黏附、生长和分化的影响**★

课题背景:寻求能够作为细胞移植与引导组织生成的人工合成可降解的生物材料作为组织工程支架的应用成为组织工程的研究重点。本课题受国家自然科学基金资助(30300084),旨在检测实验室合成的聚乙二醇接枝的聚乳酸材料(PPLA)是否具有良好的生物相容性。

应用要点:采用了小相对分子质量聚乙二醇接枝改性了聚乳酸材料,通过对牛血清白蛋白的吸附实验发现,该材料能显著地降低对非特异性蛋白的吸附;通过细胞实验发现该材料具有促进细胞黏附、生长和分化功能,加上前期研究已经证实的它的良好亲水性和降解性,因此它是一种比聚乳酸更能满足生物材料发展要求的新型材料。

同行评价:文章研究聚乙二醇接枝聚乳酸作为骨生长的生物材料介质,是热门课题,实验的结果良好,数据可靠,具有很强的应用价


摘要
目的:在前期成功合成聚乙二醇接枝的聚乳酸材料[poly(ethylene glycol) graft poly(D,L-lactic acid),PPLA]的基础上,以聚乳酸为对照,进一步考察该材料的细胞相容性。
方法:实验于2006-10在重庆大学生物材料与仿生工程实验室进行。①以FITC荧光标记牛血清白蛋白检测聚乳酸及自制的PPLA材料的抗非特异性蛋白吸附性能。②在体外聚乳酸和PPLA聚合物膜上培养Wistar新生鼠成骨细胞。采用四甲基偶氮唑盐比色法测试细胞黏附和增殖情况,并通过检测碱性磷酸酶活性评价细胞分化程度。
结果:①与聚乳酸相比,PPLA对牛血清白蛋白的吸附明显降低,仅为聚乳酸的37.3%。②与聚乳酸相比,在接种成骨细胞后,PPLA材料上的细胞黏附量更多,细胞数量更高;经过8 d的培养后,PPLA材料上成骨细胞的碱性磷酸酶活性高于聚乳酸。
结论:与聚乳酸相比,PPLA材料具有更强的抗非特异性蛋白吸附作用,并能显著地促进细胞黏附、生长及分化,具有良好细胞相容性。

Influence of poly(ethylene glycol) graft poly(D,L-lactic acid) with protein-resistant property on the adhesion, proliferation and differentiation of osteoblasts

Abstract

AIM
The poly(ethylene glycol) graft poly(D,L-lactic acid) (PPLA) has been synthesized, and this study will detect its cell compatibility compared with polylactic acid (PLA).
METHODS: The experiment was carried out in the laboratory of Biomaterials and Bionics, Chongqing University in October 2006.①The anti-non-specific protein absorption properties of PLA and PPLA polymer were tested using FITC labeled bovine serum albumin.②Osteoblasts of Wistar neonatal rats were cultured on the PLA and PPLA polymer in vitro. The adhesion and proliferation of osteoblasts on the films of polymers were tested using 3-(4, 5-dimethylthiazal-2yl) 2, 5-diphenyltetrazolium bromide assay. The alkaline phosphatase activity was tested to evaluate the cell differentiation.
RESULTS: ①Compared with PLA, the poly(ethylene glycol) graft PPLA showed an obviously decreasing absorption of bovine serum albumin, accounting for 37.3% of PLA.②Compared with PLA, the adhesion and proliferation of osteoblasts were improved after seeding on PPLA material; Alkaline phosphatase activity of osteoblasts was also advanced on PPLA material after 8 days.
CONCLUSION: Compared with PLA, PPLA reveals a prominent property of non-specific protein absorption, it can significantly improve the adhesion, proliferation and differentiation of osteoblasts, as well as representing good cell compatibility.
