Influence of human cord blood mononuclear cell transplantation on haematopoiesis and immune function in nude mice Abstract:A randomized control animal experiment was performed at the SPF Animal Laboratory of Liaoning Medical University from November 2006 to February 2007. Balb/C nu+ nude mice were radiated with 60Co before transplantation. Fresh human cord blood mononuclear cells were infused into mice in the cord blood transplantation group after irradiation, whereas mice in the control group were infused with saline. Survival rate and survival time in mice of cord blood transplantation group were significantly higher than in the control group. Human CD45+ cell of bone marrow arose in 2 weeks and rapidly increased, and kept a high level in 4-8 weeks, and reached a peak in 8 weeks, then significantly decreased, and could be detected until 12 weeks. Two weeks after transplantation, human specificity Cart-1 genes were found in bone marrow and peripheral blood of mice, and can be detected until 12 weeks. No human DNA was detected in the control group. It is confirmed that cord blood mononuclear cells can be successfully infused into immune deficiency BALB/cnu+ nude mice, which can achieve reconstitution of haematopoiesis and immune system in immune deficiency mice.
