复合成骨细胞藻酸盐凝胶的体内成骨效应**☆

课题背景:课题获得湖北省教育厅基金项目(B200524005)和十堰市科技攻关项目(2006050D)资助,本实验只是其中的阶段性成果,证实复合成骨细胞藻酸盐凝胶具有一定的异位成骨能力。
应用要点:①本实验构建的组织工程骨具有正常骨结构,可进一步完成骨组织的改建。②可注射性藻酸钙凝胶作为一种新型组织工程载体,给成骨细胞的生长增殖及代谢创造了一个良好的微环境。
偏倚或不足:实验构建的组织工程骨成骨确切、可靠,但实验过程中尚有许多问题亟待解决,如藻酸钙凝胶在体内降解过程和成骨的关系,注射该凝胶的时间、频率,最适细胞浓度等。
摘要
目的: 为使骨缺损的治疗简便可行,又要保证可靠的骨修复效果,实验将成骨细胞复合于藻酸钙水凝胶中,直接经皮注射动物体内了解其成骨情况。
方法:实验于2006-01/2007-07在十堰市太和医院生命科学研究所实验室完成。将培养的兔骨髓基质成骨细胞复合于1.5%藻酸钠溶液,再加入葡萄糖酸钙,制备成骨细胞密度为1×1010 L-1、钙离子浓度为50 mmol/L的藻酸钙水凝胶。将20只健康成年新西兰大白兔左侧背部经皮注射成骨细胞-藻酸钙水凝胶复合物(实验侧),右侧注射单纯藻酸钙水凝胶(对照侧)。于术后4,8,12周分别行X射线检查及组织学观察成骨情况。
结果:20只健康成年新西兰大白兔均进入结果分析。X射线和组织学观察表明,实验侧成骨活跃,骨再生迅速,12周内,新生骨组织明显增多,且新生骨组织趋向成熟,X线片显示高密度影;而对照侧未见骨组织出现。
结论:复合成骨细胞的藻酸钙凝胶经皮注射植入在动物体内异位成骨可靠、确切。

Osteogenesis ability of composite calcium alginate hydrogel and osteoblasts
Abstract
AIM:To explore a convenient and feasible therapy for bone defects, and ensure its reliable effect to repair bone defects, this study was designed to compound osteoblasts with calcium alginate hydrogel, and observe the osteogenesis ability in animals by percutaneous injection.
METHODS: The experiment was carried out in the laboratory of Life Sciences Institute of Shiyan Taihe Hospital between January 2006 and July 2007. New Zealand rabbit bone marrow stromal osteoblasts cultured in vitro were mixed with 1.5% sodium alginate and calcium gluconate, resulting in a cell density of 1×1010 L-1 and 50 mmol/L calcium alginate hydrogel. The mixture was injected into the dorsal subcutaneous tissue of twenty New Zealand white rabbits in the left side (experimental side); the calcium alginate hydrogel was injected into the right side of rabbits (control side). All specimens were examined with X-ray and histology on the 4, 8, 12 postoperative weeks.
RESULTS: Twenty New Zealand white rabbits were all involved in the result analysis. X-ray and histological observations showed that the bone formation of experimental side with rapid regeneration was fast 12 weeks after injection. X-ray photograph indicated high density of calcified image in the experimental side; while no bone formation was found in the control side.
CONCLUSION: Percutaneous injection of calcium alginate hydrogel and osteoblasts composites can result into satisfactory ectopic osteogenesis in animals.
